Assistant corrections commissioner Bob Wise said this afternoon that the
Melbourne underworld figure had shared his Acacia unit at Barwon jail with two other men and spent six hours a day in a day room with them.
The two men have been separated since the fatal attack.
Williams, 39, died of cardiac arrest about 1.30pm after receiving treatment for head injuries.
Williams was serving life imprisonment in Barwon's maximum security Acacia unit with a non-parole period of 35 years for ordering the murders of three underworld rivals and the failed conspiracy to murder a fourth person.He pleaded guilty in 2007 to murdering drug dealer Jason Moran and his father Lewis Moran and amphetamines trafficker Michael Marshall.
Williams was a key player in Melbourne's underworld wars, in which 35 people were killed from 1998 to 2009.
An Ambulance Victoria spokesman said attempts to revive a man who had been seriously injured after being assaulted failed and he died at the scene.
Victoria Police said in a statement they were “in the process of notifying the family and will have more details shortly”.
Homicide detectives have arrived at the prison and a crime scene has been established.
Williams had one child, a daughter, with his ex-wife Roberta Williams, a convicted drug trafficker.
The decision by Victoria Police to pay for Williams' daughter's private school fees was on the front page of The Herald Sun today.

Police said the assault occurred in an exercise yard and two other inmates and a weapon were involved.
A prison officer was about 10 metres away when the incident occurred, corrections officials said.
Corrections Minister Bob Cameron said Williams was housed in a unit with two other inmates who have been placed in a secure area.
He said the three men were allowed to associate with each other for six hours a day, between 8am and 2pm, but were not always supervised.
Mr Cameron said three investigations would be held into Williams' death. He said they would be conducted by police, the coroner and the Office of Corrective Services.
Deputy corrections commissioner Rod Wise said it was not practical to have every prisoner supervised when they were out of their cell

The jailmate of Carl Williams has today been sentenced to life behind bars for murdering the notorious gangland killer inside Barwon Prison.
In a packed Victorian Supreme Court, Justice Lex Lasry described Matthew Charles Johnson’s actions in murdering Williams on April 19 last year in the high-security Acacia unit they shared as "appalling"...
He ordered Johnson, who showed no emotion in court, to serve a minimum of 32 years before becoming eligible for parole.
